Single-Step Method of RNA Isolation by Acid Guanidinium Thiocyanate–Phenol–Chloroform Extraction

1987 · paper · Biomedicine

Describes a single-step method for isolating total RNA from tissues or cells using an acid guanidinium thiocyanate–phenol–chloroform mixture. It yields pure, undegraded RNA in about 4 hours and suits many samples or very small amounts.

1 finding

Substance Table 1 step 1 reads "Solution D, 0.2 M sodium acetate, pH 4, phenol, chloroform (1:0.1:1:0.2)", but the protocol text adds "0.1 ml of 2 M sodium acetate, pH 4" per 1 ml of solution D.
  • p. 2 Experimental procedure, left column
    0.1 ml of 2 M sodium acetate, pH 4
  • p. 2 Table 1, step 1 Extraction
    Solution D, 0.2 M sodium acetate, pH 4, phenol, chloroform (1:0.1:1:0.2)

Only the authors can say what is right here.

Judge's note: Checked both crops on page 2. The text says 2 M, and Table 1 prints 0.2 M for the same step with the same volume ratio (1 ml D : 0.1 ml acetate : 1 ml phenol : 0.2 ml chloroform). The paper gives no other variant of the protocol. One possible innocent reading is that 0.2 M is roughly the acetate concentration after mixing with solution D (2*0.1/1.1 ≈ 0.18 M). However, the other entries in that cell are stock reagents given by volume ratio, so the two statements are inconsistent as printed. Only the authors can say which value is meant; most likely the stock is 2 M.
