Bimekizumab versus Adalimumab in Plaque Psoriasis

New England Journal of Medicine, 2021 · paper · Clinical trials

A randomized trial in 478 patients with moderate-to-severe plaque psoriasis comparing bimekizumab with adalimumab. At week 16, 86.2% on bimekizumab and 47.2% on adalimumab had a 90% reduction in PASI score. Bimekizumab caused more oral candidiasis and diarrhea.

Reference or label Reference 11 reads "Reich K, Warren RB, Lebwohl M, et al. Bimekizumab versus secukinumab in plaque psoriasis. N Engl J Med 2021; 384:142-52."; the volume appears to be 385.

Judge's note: Rests on outside knowledge: the BE RADIANT paper was printed in N Engl J Med 2021;385:142-52, in the same issue as this article. Page 12 image confirms '384:142-52.' The page 1 crop shows this article as 'N Engl J Med 2021;385:130-41', so a page range of 142-52 runs straight on from it, which supports volume 385.

3 minor (typos and the like; most checked by the judge only)
Wording Table 3 footnote **: "From weeks 24 to 56, one case each of colon cancer and squamous-cell carcinoma was reported in the group receiving bimekizumab every 4 weeks and then every 8 weeks" gives no site for the SCC, yet NMSC is 0 for that column.
  • p. 9 Table 3, footnote ** and NMSC row (weeks 24-56, Q4W→Q8W column)
one case each of colon cancer and squamous-cell carcinoma was reported in the group receiving bimekizumab every 4 weeks and then every 8 weeks

Only the authors can say what is right here.

Judge's note: Page 9 image checked. For weeks 0-24 in the Q4W→Q8W column, NMSC = 3, which matches the 3 basal-cell carcinomas; the anal SCC, which has its site named, is left out. For weeks 24-56 in the same column, Cancer = 2 (1.3) and NMSC = 0, so the unqualified SCC must be non-cutaneous, but the footnote does not say so, unlike the other non-skin SCCs. This is an ambiguity that only the authors can resolve.

Typo Table 1 footnote ¶ reads "Dermatology Life Quality Assessment (DLQI)", whereas the Methods define it as "Dermatology Life Quality Index (DLQI)".
  • p. 5 Table 1, footnote ¶
Scores on the Dermatology Life Quality Assessment (DLQI) range from 0 to 30
  • p. 3 Efficacy End Points
    Dermatology Life Quality Index (DLQI)
  • p. 5 Table 1 footnote ¶
    Dermatology Life Quality Assessment (DLQI)

Judge's note: Page 5 image confirms 'Assessment' in the footnote. The page 3 crop confirms 'Dermatology Life Quality Index (DLQI)' in the Methods. The two expansions of the abbreviation do not match.

Typo Table 1 footnote ‡ reads "Scores on the Psoriasis and Severity Index (PASI)"; 'Area' is missing (should be 'Psoriasis Area and Severity Index').
  • p. 5 Table 1, footnote ‡
Scores on the Psoriasis and Severity Index (PASI) range from 0 to 72
  • p. 5 Table 1 footnote ‡
Scores on the Psoriasis and Severity Index (PASI)

Judge's note: Page 5 image checked: the footnote prints 'Psoriasis and Severity Index'. The abstract (page 1) and the Methods (page 2) both expand PASI as 'Psoriasis Area and Severity Index'.
