Paper check About

The 21-Nucleotide let-7 RNA Regulates Developmental Timing in Caenorhabditis elegans

Nature, 2000 · paper · Biomedicine

The text provided is a fragment of a different Nature letter, about sodium release by the Na+/K+ pump in squid axons, and does not show the paper itself. The let-7 paper's findings cannot be described from it.

12 minor (typos and the like; most checked by the judge only)
Wording "underwent a supernumerary moult to a fifth larval stage, L5 (56%, n = 26)": no whole number of animals out of 26 gives 56%, either by rounding or by truncation.
  • p. 2 Main text, left column, supernumerary moult
    L5 (56%, n = 26)
  • p. 2 column 2, supernumerary moult
    L5 (56%, n 26)

Only the authors can say what is right here.

Judge's note: Checked main-p002.png and the C010-a crop. The text reads '(56%, n = 26)'. 14/26 = 53.8% and 15/26 = 57.7%, so either the n or the percentage may be misprinted. 14/25 would give 56%. I found nothing in the paper that gives another denominator or says data were pooled. Only the authors can say which value is right.

Second check: another model agreed

Wording 'A reporter bearing the lin-41 3′ UTR was expressed in larval stage but not adult wild-type animals (Fig. 4b, c)' sits beside 'only 19% (n = 21) of wild-type adults' expressing the same lacZ/lin-41 3′ UTR fusion.
  • p. 5 Results, left column
expressed in larval stage but not adult wild-type animals
  • p. 5 Results, left column
    only 19% (n = 21) of wild-type adults

Only the authors can say what is right here.

Judge's note: Page 5 image, left column, third paragraph. The categorical 'not adult wild-type animals' is followed three sentences later by the quantification that 19% of wild-type adults (n = 21) expressed the reporter, against 79% of let-7(n2853) adults. Expression was reduced, not absent. The first sentence may describe the representative images in Fig. 4b, c, but as worded it overstates. Authors could write 'rarely expressed' or similar.

Second check: another model agreed

Typo Table 1, lin-42(n1089); let-7(mn112) unc-3(e151), L3 moult: "13 (66)". No count out of 66 rounds to 13% (9/66 = 13.6%). Only truncation gives 13, but the rest of the table rounds normally.
  • p. 2 Table 1, lin-42(n1089); let-7(mn112) unc-3(e151), L3 moult
    13 (66)
  • p. 2 Table 1, lin-42(n1089); let-7(mn112) unc-3(e151), L3 moult
    13

Judge's note: Checked the table in main-p002.png and the C010-a crop. The row reads 13 (66). Elsewhere the table and footnote round normally: 70% for 32/46 = 69.6, 7% for 4/60 = 6.7, 28% for 20/72 = 27.8. So 13% fits truncation of 9/66, not the table's rounding. This is a minor point.

Typo Table 1, lin-14(n179) let-7(mn112) unc-3(e151) 25 °C, L3 moult: "18 (32)". 6/32 = 18.75%, which rounds to 19%, and 5/32 = 15.6%. Only truncation gives 18, but the rest of the table rounds normally.
  • p. 2 Table 1, lin-14(n179) let-7(mn112) unc-3(e151) 25 °C, L3 moult
    18 (32)
  • p. 2 Table 1, lin-14(n179) let-7(mn112) unc-3(e151) 25 °C, L3 moult
    18

Judge's note: Checked the table in main-p002.png. The row reads 18 (32) 30 (129). The other entries round normally (70% for 32/46, 7% for 4/60), so 18% does not follow the table's rounding. This is a minor point.

Typo "the strongest retarded heterochronic defects in a lin14(+) background": the gene name is missing its hyphen and should read lin-14(+).
  • p. 2 Main text, left column, genetic screen paragraph
    lin14(+)
  • p. 2 Results, para 2
retarded heterochronic defects in a lin14(+) back- ground (Fig. 1c; Table 1)
  • p. 2 Figure 1 legend
c, let- 7(n2853) L4 stage animal with LIN-29 expression reduced in V cells
  • p. 2 paragraph 2
    in a lin14(+) back-

Judge's note: Checked main-p002.png, left column. 'lin14(+)' is printed without the hyphen, while the gene is written lin-14 everywhere else. The reviewers' separate point about the Fig. 1c pointer is judged under C009.

Typo Table 1 genotype notation is inconsistent. The row "lin-28(n719); let-7(mn112); unc-3(e151)‡" has a semicolon between let-7 and unc-3, although both genes are on LG X. The footnote writes "lin-28 n719" without parentheses and "lin-14(n536n540)" without the space used in the table.
  • p. 2 Table 1, strain column
    lin-28(n719); let-7(mn112); unc-3(e151)‡
  • p. 2 Table 1 footnote ²
    lin-28 n719; let-
  • p. 2 Table 1 footnote ²
    lin-14(n536n540)

Judge's note: Checked main-p002.png. Every other row, the footnote's own 7% entry, the Fig. 3 legend and the Methods write 'let-7(mn112) unc-3(e151)' with no semicolon. Fig. 2a shows let-7 and unc-3 together on LG X. The footnote shows the other two inconsistencies. The meaning is clear, so this is cosmetic.

Typo Inconsistent gene-name forms: 'downregulation of a lac-Z reporter gene' (p. 5) versus 'lacZ' elsewhere (e.g. 'The lacZ/lin-41 3′ UTR', 'col-10/lacZ'), and 'in a lin14(+) background' (p. 2) versus 'lin-14' elsewhere.
  • p. 5 Main text, left column
    lac-Z reporter gene
  • p. 5 Results, left column
    a lac-Z reporter gene
  • p. 2 Results, para 2
    lin14(+) back-
  • p. 5 paragraph 3
    a lac-Z

Judge's note: Page 5 image, left column, third paragraph: 'lac-Z' ends a line but the Z stays on the same line, so the hyphen is printed and is not a line-break hyphen. A few lines later the same sentence context has 'lacZ/lin-41', and the Methods have 'col-10/lacZ'. The page 2 image, left column, shows 'lin14(+) back-' with no hyphen, while the gene is written lin-14 everywhere else. Both are cosmetic.

Typo "three of the four lin-14 elements are conserved in other Caenorhabiditae": the taxon name is misspelled (should read Caenorhabditidae).
  • p. 4 Main text, right column, end of first paragraph
    conserved in other Caenorhabiditae
  • p. 4 Results, last para before Fig. 4
    conserved in other Caenorhabiditae.

Judge's note: Checked main-p004.png, right column. The word is printed 'Caenorhabiditae' on the page.

Typo Fig. 4 legend: "caused a bursting phenotype similar to let-7(N2853) (g)". The allele should be lower case, n2853.
  • p. 4 Figure 4 legend, panels f, g
    similar to let-7(N2853) (g)
  • p. 4 Figure 4 legend, last sentence
    similar to let-7(N2853)
  • p. 4
caused a bursting phenotype similar to let-7(N2853) (g).

Judge's note: Checked main-p004.png. The legend prints 'N2853' with a capital N. The panel g label and the rest of the paper use n2853.

Typo Fig. 4 legend: "(between asterisks in a allows expression in let-7(+) adults (e)". The parenthesis opened before 'between' is never closed.
  • p. 4 Figure 4 legend, panel e
(between asterisks in a allows expression
  • p. 4 Figure 4 legend
(between asterisks in a allows expression in let-7(+) adults (e)
  • p. 4 Figure 4 legend, b–e
    (between asterisks in a allows

Judge's note: Checked main-p004.png. No closing parenthesis follows 'in a'.

Typo 'complementary sites in the 3′ UTRS of lin-42 or daf-12' uses a capital S; the plural is written 'UTRs' elsewhere.
  • p. 5 Main text, left column
    3′ UTRS of lin-42 or daf-12
  • p. 5 paragraph 3, last sentence
    39 UTRS of lin-42 or daf-12

Judge's note: Page 5 image, left column, end of the third paragraph, clearly shows 'UTRS'. Elsewhere the paper writes 'UTRs' (e.g. '26 3′ UTRs' in the first paragraph of the same page).

Second check: none (judge only)

Typo Reference 12 prints the first author as 'Zucker-Sparison, E. & Blumenthal, T.'
  • p. 5 Reference 12
    Zucker-Sparison, E. & Blumenthal, T.
  • p. 5 reference 12
    Zucker-Sparison, E.

Only the authors can say what is right here.

Judge's note: Rests on outside knowledge: the first author of J. Mol. Evol. 28, 487–496 (1989) is E. Zucker-Aprison. The page 5 reference list shows 'Zucker-Sparison' exactly as quoted. The correct spelling cannot be checked from the files.
