Induced Pluripotent Stem Cell Lines Derived from Human Somatic Cells

2007 · paper · Biomedicine

Shows that four factors, OCT4, SOX2, NANOG and LIN28, can reprogram human somatic cells into pluripotent stem cells. The resulting cells have normal karyotypes, express embryonic stem cell markers and telomerase, and can differentiate into derivatives of all three germ layers.

Reference or label The Fig. 3 legend for panel B (quantitative RT-PCR of OCT4 and NANOG) ends "The data are presented as mean ± SD from three telomerase activity assays.", although the panel shows no telomerase data.
  • p. 3 Fig. 3 legend, last sentence (panel B)
The data are pre- sented as mean ± SD from three telomerase activity assays.
  • p. 3 Fig. 2 legend, panel B
presented as mean ± SD from three telomer- ase activity assays
  • p. 3 Fig. 3 legend, last sentence (panel B)
sented as mean ± SD from three telomerase activity assays.
  • p. 3 Fig. 3 legend, end of (B)
mean ± SD from three telomerase activity assays

Only the authors can say what is right here.

Judge's note: On the page 3 image, Fig. 3A is a 1-PCC clustering dendrogram and Fig. 3B plots "Relative expression" of OCT4 and NANOG (Total/Endogenous) by quantitative RT-PCR. Telomerase activity appears only in Fig. 2B, whose legend carries the identical sentence, where it fits. The Fig. 3 sentence therefore mislabels the source of the error bars. The actual number and kind of replicates behind Fig. 3B is not stated anywhere in the article, so the authors must supply the correct wording.

3 minor (typos and the like; most checked by the judge only)
Typo The same factor is printed as italic lower-case "c-myc" in "factors (Oct4, Sox2, c-myc, and Klf4) were shown" and as roman "c-Myc" a few lines later in "of c-Myc causes death and differentiation of human ES cells".
  • p. 2 Introduction, first paragraph
    factors (Oct4, Sox2, c-myc, and Klf4) were shown
  • p. 2 Introduction, first paragraph
of c-Myc causes death and differentiation of hu- man ES cells

Judge's note: Both crops show the wording as quoted: italic "c-myc" sits in a list with capitalised Oct4, Sox2 and Klf4, and roman "c-Myc" follows in the same paragraph, where it is then called "this gene". Italic lower-case c-myc was a common way to write the mouse gene, so either form could be legitimate; this is a minor capitalisation inconsistency only.

Typo The Fig. 3 legend reads "“Endogenous” indicates that primers were included in the 3′ untranslated region measure expression of the endogenous gene only", which is ungrammatical (two finite verbs with no connector).
  • p. 3 Fig. 3 legend, panel B definition of "Endogenous"
indicates that primers were included in the 3′ untranslated re- gion measure expression of the en- dogenous gene only
  • p. 3 Fig. 3 legend
included in the 3′ untranslated region measure expression of the endogenous gene only

Judge's note: The crop confirms the wording as printed. The parallel clause that follows ("primers in coding regions measure expression of both the endogenous gene and the transgene") shows the intended sense: "primers in the 3′ untranslated region measure expression of the endogenous gene only". Either "were included" is stray or a "to" is missing before "measure". This is a copy-editing slip and the meaning is recoverable.

Typo A cell line is named "ITT026" (digit zero) in "Lines designated as ITT026 and ITT4 iPS were randomly chosen" and "ITTO26" (letter O) in "fibroblast-derived ITTO26 and ITT4 iPS cells" on PDF page 5.
  • p. 5
    fibroblast-derived ITTO26 and ITT4 iPS cells

Judge's note: The text layer has "ITT026" at line 772 and "ITTO26" at line 783 of extract/main.txt. In the crop, the fourth character of the second occurrence is a round glyph wider than the adjacent digits, consistent with a letter O. I did not view the first occurrence at high resolution; the full-page image is too coarse to tell the glyphs apart, so that side rests on the text layer. Caveat: both occurrences are in the next article that begins on PDF page 5 (Hanna et al., "Treatment of Sickle Cell Anemia Mouse Model with iPS Cells Generated from Autologous Skin"), not in the paper under review, whose text ends with the references at the top of that page. It is recorded as an observation only because page 5 is listed as in scope.
