A Programmable Dual-RNA-Guided DNA Endonuclease in Adaptive Bacterial Immunity

Science, 2012 · paper · Biomedicine

Shows that in some bacterial CRISPR systems, a crRNA paired with a tracrRNA directs the Cas9 protein to cut both strands of target DNA. A single engineered RNA chimera works too, suggesting use for RNA-programmable genome editing.

6 minor (typos and the like; most checked by the judge only)
Reference or label Reference 36 gives a year that does not match the cited volume: "Miller JC et al., Nat Biotechnol 29, 143–148 (2010). [PubMed: 21179091]".
  • p. 8 Reference 36
Miller JC et al., Nat Biotechnol 29, 143–148 (2010)

Only the authors can say what is right here.

Judge's note: Rests on outside knowledge: Nature Biotechnology volume 29 (pages 143-148, PMID 21179091) is the 2011 issue, although that article appeared online in late December 2010. Opened crop C009-a (p. 8) and confirmed the entry prints '29, 143-148 (2010)'. Neighbouring entries in the same list cite the issue year (e.g. ref. 35 Genetics 186 (2010), ref. 38 Genetics 188 (2011)), so an advance-online date is a possible but inconsistent explanation; only the authors can say which they intended. Severity is a reference-detail one.

Typo Correspondence footnote on p. 1 has an unclosed parenthesis: "Correspondence should be addressed to J.A.D. (doudna@berkeley.edu) and E.C. (emmanuelle.charpentier@mims.umu.se."
  • p. 1 Footnote, correspondence line
    and E.C. (emmanuelle.charpentier@mims.umu.se.

Judge's note: Opened crop C001-a (and the p.1 full page): the typeset footnote shows the opening bracket before the second address and no closing bracket, the line ending '...mims.umu.se.' The parallel first address is correctly closed as '(doudna@berkeley.edu)'. Not an extraction artefact, since the rendered page shows the same.

Typo The first author's surname is set two ways in the same document: "Martin Jinek" in the byline and running head, but "Jínek M" in references 6 and 11.
  • p. 1 Author byline
    Martin Jinek
  • p. 7 References and Notes, ref. 6
Haurwitz RE, Jínek M, Wiedenheft B, Zhou K, Doudna JA
  • p. 7 References and Notes, ref. 11
Sashital DG, Jínek M, Doudna JA

Judge's note: Opened crops C002-a (byline: 'Martin Jinek#1,2'), C002-b (ref. 6 'Haurwitz RE, Jínek M, Wiedenheft B, Zhou K, Doudna JA') and C002-c (ref. 11 'Sashital DG, Jínek M, Doudna JA'). The acute accent is clearly rendered in the typeset reference list and absent in the byline and in the 'Jinek et al.' running heads, so the inconsistency is in the published pages, not in the text layer. Cosmetic only.

Typo Missing word space on p. 6: "This single transcript effectively fuses the 3’end of crRNA to the 5’ end of tracrRNA".
  • p. 6 Results, 'Cas9 can be programmed with a single chimeric RNA' section
This single transcript effectively fuses the 3’end of crRNA to the 5’ end of tracrRNA

Judge's note: Opened crop C005-a: the typeset line reads '3’end of crRNA' with no space, while '5’ end of tracrRNA' in the same line clearly has one, and the Fig. 5A legend (p. 14) writes 'the 3’ end of crRNA to the 5’ end of tracrRNA' with spaces. Cosmetic typesetting slip.

Second check: none (judge only)

Typo Hyphen misplaced in the expansion of TALEN: "Transcription-Activator Like Effector Nucleases (TALENs)" (p. 6, Conclusions).
  • p. 6 Conclusions
    Transcription-Activator Like Effector Nucleases (TALENs)

Only the authors can say what is right here.

Judge's note: Rests on outside knowledge: the conventional expansion of TALEN is 'transcription activator-like effector nuclease', with the hyphen in 'activator-like'. Opened crops C007-a and C006-a: the page plainly prints 'Zinc-Finger Nucleases (ZFNs) and Transcription-Activator Like Effector Nucleases (TALENs)', so the hyphen is present but after 'Transcription'. Nothing inside the paper contradicts the printed form, and the acronym is unaffected, so this is cosmetic only.

Typo Missing terminal period in the Acknowledgements: "financial support to K.C and hosting K.C. in their laboratories at MFPL".
  • p. 7 Acknowledgements, final sentences
financial support to K.C and hosting K.C. in their laboratories at MFPL

Judge's note: Opened crop C008-a (p. 7): the typeset line shows 'financial support to K.C and hosting K.C. in their laboratories at MFPL', the first instance lacking the final point. Every other occurrence in the same paragraph ('K.C. and E.C.', '(K.C.)', 'K.C. is a fellow') carries it. Cosmetic.
